Electroencephalographic slow wave activity (SWA) during slow wave sleep (SWS) undergoes dynamic fluctuations in reaction to sleep/wake history. SWA increases as a consequence of prior waking and decreases as consequence of prior SWS. These fluctuations are evidence for a homeostatic regulatory process, the neurobiological underpinnings of which remain to be defined. The anti-neuroinflammatory agent minocycline abolishes the increase in SWA that normally occurs after 1- or 3-h sleep deprivation. We sought to determine whether this effect is also observed during spontaneous sleep. We describe a novel procedure for measuring the predictive relationship between spontaneous changes in sleep/wake states in the short-term (less than 30 min) and subsequent SWA. In saline-treated mice, 16 or more minutes of spontaneous wakefulness during a 20-min interval causes an increase in SWA during subsequent SWS, and 16 or more minutes spent in SWS causes a decrease in SWA during subsequent SWS. Minocycline administration (45 mg/kg) abolishes the increase caused by wakefulness but not the decrease caused by sleep. These data demonstrate that minocycline attenuates SWA dynamics in spontaneous sleep. Inflammatory events in the brain may underlie, in part, wakefulness-induced changes in the sleep electroencephalogram.

Die Anti-neuroinflammatorische Agent Minocyclin wird abgeschafft, die Zunahme der SWA, das normalerweise nach 1 - oder 3-h Entbehrung schlafen auftritt. Wollten wir bestimmen, ob dieser Effekt auch w&auml;hrend der spontanen Schlaf beobachtet wird. Kochsalzl&ouml;sung-behandelten M&auml;usen 16 oder mehr Minuten spontane Wachheit w&auml;hrend einer 20-Minuten-Intervall bewirkt eine Erh&ouml;hung in SWA w&auml;hrend nachfolgende SWS und mindestens 16 Minuten verbrachte in SWS bewirkt eine Abnahme der SWA w&auml;hrend der nachfolgenden SWS. Minocyclin Verwaltung (45 mg/kg) wird abgeschafft, die Erh&ouml;hung durch Wachheit verursacht, aber nicht die Abnahme durch Schlaf verursacht. Methamphetamine and modafinil exert their wake-promoting effects by elevating monoaminergic tone. The severity of hypersomnolence that occurs subsequent to induced wakefulness differs between these two agents. Microglia detects and modulates CNS reactions to agents such as D-methamphetamine that induce cellular stress. We therefore hypothesized that changes in the sleep/wake cycle that occur subsequent to administration of D-methamphetamine are modulated by cerebral microglia. In CD11b-herpes thymidine kinase transgenic mice (CD11b-TK(mt-30)), activation of the inducible transgene by intracerebroventricular (icv) ganciclovir results in toxicity to CD11b-positive cells (i.e. microglia), thereby reducing cerebral microglial cell counts. CD11b-TK(mt-30)and wild type mice were subjected to chronic icv ganciclovir or vehicle administration with subcutaneous mini-osmotic pumps. D-methamphetamine (1 and 2 mg/kg), modafinil (30 and 100 mg/kg) and vehicle were administered intraperitoneally to these animals. In CD11b-TK(mt-30) mice, but not wild type, icv infusion of ganciclovir reduced the duration of wake produced by D-methamphetamine at 2 mg/kg by nearly 1h. Nitric oxide synthase (NOS) activity, studied ex vivo, and NOS expression were elevated in CD11b-positive cerebral microglia from wild type mice acutely exposed to d-methamphetamine. Additionally, CD11b-positive microglia, but not other cerebral cell populations, exhibited changes in sleep-regulatory cytokine expression in response to d-METH. Finally, CD11b-positive microglia exposed to d-methamphetamine in vitro exhibited increased NOS activity relative to pharmacologically-naÃ¯ve cells. CD11b-positive microglia from the brains of neuronal NOS (nNOS)-knockout mice failed to exhibit this effect. We propose that the effects of D-METH on sleep/wake cycles are mediated in part by actions on microglia, including possibly nNOS activity and cytokine synthesis.

STUDY OBJECTIVES: Sleep loss triggers changes in inflammatory signaling pathways in the brain and periphery. The mechanisms that underlie these changes are ill-defined. The Toll-like receptor 4 (TLR4) activates inflammatory signaling cascades in response to endogenous and pathogen-associated ligands known to be elevated in association with sleep loss. TLR4 is therefore a possible mediator of some of the inflammation-related effects of sleep loss. Here we describe the baseline electroencephalographic sleep phenotype and the biochemical and electroencephalographic responses to sleep loss in TLR4-deficient mice. DESIGN, MEASUREMENTS AND RESULTS: TLR4-deficient mice and wild type controls were subjected to electroencephalographic and electromyographic recordings during spontaneous sleep/wake cycles and during and after sleep restriction sessions of 3, 6, and 24-h duration, during which sleep was disrupted by an automated sleep restriction system. Relative to wild type control mice, TLR4-deficient mice exhibited an increase in the duration of the primary daily waking bout occurring at dark onset in a light/dark cycle. The amount of time spent in non-rapid eye movement sleep by TLR4-deficient mice was reduced in proportion to increased wakefulness in the hours immediately after dark onset. Subsequent to sleep restriction, EEG measures of increased sleep drive were attenuated in TLR4-deficient mice relative to wild-type mice. TLR4 was enriched 10-fold in brain cells positive for the cell surface marker CD11b (cells of the monocyte lineage) relative to CD11b-negative cells in wild type mouse brains. To assess whether this population was affected selectively by TLR4 knockout, flow cytometry was used to count F4/80- and CD45-positive cells in the brains of sleep deprived and time of day control mice. While wild-type mice exhibited a significant reduction in the number of CD11b-positive cells in the brain after 24-h sleep restriction, TLR4-deficient mice did not. CONCLUSION: These data demonstrate that innate immune signaling pathways active in the monocyte lineage, including presumably microglia, detect and mediate in part the cerebral reaction to sleep loss.

Ziele der Studie: Schlafmangel l&ouml;st Ver&auml;nderungen inflammatorische Signalwege im Gehirn und Peripherie. Der Toll-Like-Rezeptor 4 (TLR4) aktiviert entz&uuml;ndliche Signalling Kaskaden als Reaktion auf endogene und Pathogen-assoziierte Liganden, die bekannterma&szlig;en mit Schlafmangel erh&ouml;ht. TLR4 ist daher eine m&ouml;gliche Vermittler von einige der Entz&uuml;ndung-bezogenen Auswirkungen von Schlafmangel. Hier beschreiben wir die Grundlinie EEG Schlaf Ph&auml;notyp und die biochemischen und EEG Reaktionen auf Verlust TLR4-defizienten M&auml;usen zu schlafen. DESIGN, Messungen und Ergebnisse: TLR4-defizienten M&auml;usen und Wildtyp-Steuerelemente wurden unterworfen EEG und elektromyographische Aufnahmen w&auml;hrend spontaner Schlaf/Wach-Zyklen und w&auml;hrend und nach Schlaf Beschr&auml;nkung Sitzungen 3, 6 und 24-h Dauer, w&auml;hrend der Schlaf, durch eine automatisierte Schlafsystem Beschr&auml;nkung gest&ouml;rt wurde. Vergleich zu Wildtyp-Steuerelement M&auml;use, ausgestellt TLR4-defizienten M&auml;usen eine Zunahme der Dauer der prim&auml;ren t&auml;glich Erwachen Anfall auftreten zu dunklen Beginn in einem hell-dunkel-Zyklus. Die aufgewendeten Zeit im nicht-REM-Schlaf von TLR4-defizienten M&auml;usen verringerte sich proportional erh&ouml;hte Wachheit in den Stunden unmittelbar nach dunklen auftreten. Im Anschluss an die Schlaf-Beschr&auml;nkung wurden EEG Ma&szlig;nahmen erh&ouml;hte Schlaf-Laufwerks TLR4-defizienten M&auml;usen im Vergleich zu Wildtyp M&auml;usen abgeschw&auml;cht. TLR4 war angereicherten 10fach verbesserter im Gehirnzellen positiv f&uuml;r die Zelle Oberfl&auml;che Marker CD11b der Monozyt Linie relativ CD11b-Negative Zellen im Gehirn der Wildtyp-Maus mit. Um zu beurteilen, ob diese Population von TLR4-ko selektiv betroffen war, Durchflusszytometrie wurde verwendet, um die F4/80 z&auml;hlen- und CD45-positiven Zellen in den Gehirnen der Schlaf entzogen und die Uhrzeit des Tages Kontrolle M&auml;use. Qui descriviamo il fenotipo di sonno elettroencefalografici baseline e le risposte biochimiche ed elettroencefalografici a dormire perdita nei topi TLR4-carenti. Successivamente restrizione di sonno, misure di EEG dell&#39;unit&agrave; maggiore sonno erano attenuati nei topi TLR4-carenti rispetto a topi wild-type. TLR4 &egrave; stato arricchito 10 volte in cellule cerebrali positive per il marcatore di superficie delle cellule CD11b (cellule del lignaggio monociti) rispetto al CD11b-negativo delle cellule nel cervello del mouse di tipo selvatico. Non-rapid eye movement sleep (NREMS) onset is characterized by a reduction in cerebral metabolism and an increase in slow waves, 1-4-Hz oscillations between relatively depolarized and hyperpolarized states in the cerebral cortex. The metabolic consequences of slow-wave activity (SWA) at the cellular level remain uncertain. We sought to determine whether SWA modulates the rate of glycolysis within the cerebral cortex. The real-time measurement of lactate concentration in the mouse cerebral cortex demonstrates that it increases during enforced wakefulness. In spontaneous sleep/wake cycles, lactate concentration builds during wakefulness and rapid eye movement sleep and declines during NREMS. The rate at which lactate concentration declines during NREMS is proportional to the magnitude of electroencephalographic (EEG) activity at frequencies of <10 Hz. The induction of 1-Hz oscillations, but not 10-Hz oscillations, in the electroencephalogram by optogenetic stimulation of cortical pyramidal cells during wakefulness triggers a decline in lactate concentration. We conclude that cerebral SWA promotes a decline in the rate of glycolysis in the cerebral cortex. These results demonstrate a cellular energetic function for sleep SWA, which may contribute to its restorative effects on brain function.

Inicio del sue&ntilde;o (NREMS) movimiento de ojo r&aacute;pido no se caracteriza por una disminuci&oacute;n del metabolismo cerebral y un aumento en las ondas lentas, 1-4-Hz oscilaciones entre Estados relativamente despolarizados y hyperpolarized en la corteza cerebral. Siguen las consecuencias metab&oacute;licas de la actividad de onda lenta (SWA) a nivel celular. Intentamos determinar si SWA modula la velocidad de la gluc&oacute;lisis dentro de la corteza cerebral. La medici&oacute;n en tiempo real de la concentraci&oacute;n de lactato en la corteza cerebral de rat&oacute;n demuestra que aumenta durante la vigilia forzada. En los ciclos de sue&ntilde;o/vigilia espont&aacute;nea, concentraci&oacute;n de lactato se construye durante la vigilia y el sue&ntilde;o de movimientos oculares r&aacute;pidos y disminuye durante la NREMS. La tasa en que lactato concentraci&oacute;n disminuye durante la NREMS es proporcional a la magnitud de la actividad electroencefalogr&aacute;fica (EEG) frecuencias de &lt; 10 Hz. La inducci&oacute;n de oscilaciones de 1 Hz, pero no 10-Hz oscilaciones, en el electroencefalograma por estimulaci&oacute;n de la optogenetic de las c&eacute;lulas piramidales corticales durante wakefulness provoca una disminuci&oacute;n en la concentraci&oacute;n de lactato. Concluimos que la SWA cerebral promueve una disminuci&oacute;n de la tasa de la glicolisis en la corteza cerebral. Estos resultados demuestran una funci&oacute;n energ&eacute;tica celular para dormir SWA, que puede contribuir a sus efectos restaurativos en la funci&oacute;n cerebral.
