1. Equipment Preparation&#160;
<ol>
	<li>Examine the ultrasound transducer for defects. Depending on the equipment used, this step may be unnecessary.
	<ol>
		<li>If an air bubble is observed, remove the screw located on the right side of the transducer head, and add sterile water through the hole with a 26 G needle. Air bubbles inside the transducer head are common. They will impede the acquisition of quality images.</li>
		<li>Check the membrane covering the probe for leaks or holes. Replace if necessary.</li>
	</ol>
	</li>
	<li>Open the software and initialize the probe.
	<ol>
		<li>Choose the cardiac package from the drop down menu, along with the appropriate transducer. Click &#34;initialize&#34;. Use a 20-60 MHz probe for mice under 35 g and a 15-45 MHz probe for mice greater than 35 g.</li>
		<li>Select the operator, animal &#38; date on the demographics screen, and select &#34;start&#34;.</li>
	</ol>

</ol>
2. Mouse Preparation Including Anesthesia, Hair Removal, and Positioning&#160;
<ol>
	<li>Anesthesia: place the mouse in an induction chamber and anesthetize with a portable, table top anesthesia machine containing an isoflurane vaporizer and a waste gas container.
	<ol>
		<li>Set the vaporizer to 3% with an oxygen flow rate of 3 L/min. This relatively high anesthesia rate is used to achieve rapid anesthetic effect and therefore minimize stress response that may influence cardiac function. &#160;The short duration of the protocol minimizes any potential risk to the animal. &#160;It is very important to always keep the anesthesia rate the same. This protocol has been optimized for exclusive use of isoflurane as the anesthetic, therefore optimal conditions for other agents may vary from this protocol. A lower depth of anesthesia may be chosen, depending on experimental needs, but once an anesthetic protocol is established, it must not be changed. Anesthesia affects heart rate and other hemodynamic measurements. Therefore, if the depth of anesthesia is changed during an experiment, the data may not be useful for analyses. If several mice are to be imaged in one day, anesthetize them separately.</li>
		<li>Monitor room temperature to ensure it is the same between experimental groups. Room temperature can affect vasoreactivity, even when the mouse is on a heated table, so it should be monitored and kept the same between experimental groups that will be compared.&#160;Although this protocol does not measure animal temperature directly, constant ambient and table temperature ensures that there is little variation in temperature between experimental groups.</li>
	</ol>
	</li>
	<li>Hair removal: Remove hair from the chest with a depilatory cream after the mouse is anesthetized. Begin application with a cotton-tipped applicator, at the clavicles, and continue to just below the diaphragm.
	<ol>
		<li>Place the mouse back in the anesthesia chamber for 1 min to allow the depilatory to work. To determine if anesthesia is effective, firmly press your thumbnail against one of the mouse&#39;s paws. If there is no withdraw, anesthesia is adequate. If the limb withdraws, place the mouse back in the anesthesia chamber for another minute.</li>
		<li>Apply a small amount of lubricating ointment to the mouse&#39;s eyes to avoid damage to the cornea.</li>
		<li>Remove hair from the chest with a 2 in x 2 in gauze pad. The chemical used in the depilatory cream is caustic, and will harm skin if it is left on for too long, so care should be taken to remove all product from the skin.</li>
		<li>Apply a skin moisturizer following hair removal.</li>
	</ol>
	</li>
	<li>Positioning: Place the mouse in a ventrodorsal position on a heated table set to 37 &#176;C. Correct positioning is imperative to the acquisition of quality images. Use a table that can capture body temperature, respiration, and heart rate. Use of an integrated rail system allows for precise positioning and subsequently, image optimization.
	<ol>
		<li>Gently tape down all four paws and apply a dime size amount of transduction gel to the chest.</li>
	</ol>
	</li>
</ol>
3. Acquisition of Images: Imaging in Parasternal Long Axis View&#160;
<ol>
	<li>Lock the ultrasound transducer into place inside the mount on the rail system, and rotate it 10&#176; counterclockwise, so that the metal probe of the transducer is positioned directly over the heart. More specifically, the probe should be on the left side of the chest, in the 2nd or 3rd intercostal space, and lateral to the sternum.
	<ol>
		<li>Manipulate the x-&#160;and y-axes located on the rail system, until the correct view is obtained.</li>
		<li>Select &#34;B Mode&#34;. This is found in the upper right portion of the system console, in order to project a 2D live image.</li>
		<li>View the following anatomic structures on the monitor:
		<ol>
			<li>The entire heart from apex to aorta - The apex will be visualized on the far left of the screen, and the aorta on the far right.</li>
			<li>The lumen of the left ventricle (LV)</li>
			<li>Posterior wall of the left ventricle (LPW)</li>
			<li>Interventricular septum (IVS)</li>
			<li>The lumen of the right ventricle (RV)</li>
			<li>Anterior and posterior mitral valve leaflets (AML &#38; PML)</li>
			<li>Ascending aorta (AO)</li>
			<li>Left atrium (LA)</li>
		</ol>
		</li>
	</ol>
	</li>
	<li>Obtain one diametric measurement of the aorta in this view by pressing the scan/freeze button to &#34;freeze&#34; the image. Then use the mouse to pull back through the video loop at the bottom of the image until the left ventricle is in systole, and the aorta is at its greatest diameter.

		<li>Click the measurement tool in the upper left corner of the screen and select the icon that looks like a diagonal line. Left click your mouse and draw a straight line from the anterior to the posterior wall of the aorta, perpendicular to its long axis. Save by pressing the &#34;Frame Store&#34; button.</li>
		<li>Create a video loop by pressing &#34;Cine Store&#34;</li>
	</ol>

4. Acquisition of Images: Imaging in Parasternal Short&#160;Axis View
<ol>
	<li>Reposition the transducer to the 3 and&#160;9 o&#39;clock positions (transverse). Angle slightly caudally by manipulating the transducer mount to achieve the best view of the aorta and LV lumen. The metal probe will be positioned horizontally and directly over the sternum.
	<ol>
		<li>Manipulate the x-&#160;and y-axes on the rail system until the correct view is obtained. The LV lumen will be seen, along with the anterolateral and posteromedial papillary muscles, which are visible on the right of the monitor. This is the standard reference point for the short axis, indicating the mid portion of the left ventricle, where dimension measurements are made. Deviating slightly from the reference point with the x-&#160;and y-axes to bring different anatomical structures in view will be necessary, but positioning is explained by referencing the above view.</li>
	</ol>
	</li>
	<li>Obtain the following measurements in the short axis view:
	<ol>
		<li>B mode 
		<ol>
			<li>Two more diametric measurements of the aorta.</li>
			<li>Three measurements of the pulmonary outflow tract.</li>

		<li>Manipulate the Y axis cranially from the papillary muscle view, until the semilunar valve of the aorta comes into focus.</li>
		<li>Obtain measurements of the aorta just above the valve at the greatest diameter.</li>
		<li>Click the measurement tool in the upper left corner of the screen, and select the icon that looks like a diagonal line.</li>
		<li>Left click your mouse and draw a straight line from the anterior to the posterior wall of the aorta.</li>
		<li>Manipulate the x-&#160;and y-axes until the main pulmonary artery bifurcates. This structure will be seen anteriorly, and to the right of the aorta on the monitor.</li>
		<li>Manipulate the y-axis cranially until the annulus of the main pulmonary artery comes into view. It will not be as clearly defined as the aorta.</li>
		<li>&#34;Freeze&#34; the image and obtain the measurement in systole.</li>
		<li>Collect three measurements in total.</li>
	</ol>
	</li>
	<li>Pulsed wave (PW) Doppler measurements: PW Mode is primarily used for hemodynamic assessment of blood flow through arteries and veins. In this protocol, it will be used to retrieve three velocity time integral measurements of the aorta and pulmonary artery.
	<ol>
		<li>Bring the aorta back into view as described in step 4.3.1, and bullet point 1.
		<ol>
			<li>Select &#34;PW Mode&#34;. This is located on the upper right corner of the system console and will produce a Doppler reading of blood flow through the aorta.</li>
			<li>Place the sample volume just above the level of the aortic valve. The x-&#160;and y-axes may need to be adjusted slightly to obtain sufficient Doppler envelopes. The envelopes should have white borders, and a hollow inside indicating laminar blood flow.</li>
			<li>Once a sufficient view is obtained, &#34;freeze&#34; the image and trace the border of the Doppler envelope. This will calculate the VTI.</li>
			<li>Rotate the &#34;Angle&#34; knob located on the system console clockwise until the segmented yellow line seen in the image on the upper right-hand side of the monitor is at 0&#176;. This yellow line represents the direction of blood flow through the vessel. Since the transducer itself is angled in such a way to produce a cross-sectional or transverse view of the heart, the line must be adjusted to 0&#176;, to align with the vertical flow of blood through the ascending aorta.</li>
		</ol>
		</li>
		<li>Place the sample volume proximal to the level of the pulmonary valve in the center of the right ventricular outflow tract and repeat VTI measurements as above. The flow of blood should appear inverted, or opposite the flow of blood in relation to the aorta on the monitor.</li>
	</ol>
	</li>
	<li>M mode measurements: M-mode imaging provides high temporal resolution of tissue motion along a single ultrasound beam, and is used to quantify cavity dimensions, as well to study valvular, myocardial, and vessel wall movement.
	<ol>
		<li>Resume &#34;B Mode&#34; and reposition the transducer to obtain the &#34;reference view&#34; (the cross sectional view of the left ventricle at the level of the papillary muscles).</li>
		<li>Press &#34;M-mode&#34;. This will produce a continuous video feed in which the motion of the following anatomical structures will be visible as a &#34;ribbon&#34;. If dilated, the RV lumen will appear at the top of the feed as a very thin black ribbon. The interventricular septum (IVS) will be visible as an opaque ribbon directly below the RV lumen. The LV lumen will be seen directly below the IVS. It is the large black space that occupies the majority of the feed. Below the LV lumen is the LV posterior wall (LVPW) which will be seen as an opaque ribbon.</li>
		<li>Freeze the image and pull back through the video loop if needed to a point where respiration is not occurring. When the mouse respires, the image acquisition is disrupted by movement of the diaphragm and chest wall, thereby producing a distorted &#34;smeared&#34; artifact in the feed that occurs with regular frequency.</li>
		<li>Obtain the following measurements using the diagonal line icon:
		<ol>
			<li>Three measurements of LV end diastolic dimension, which appears as the largest distance between the IVS and LVPW.</li>
			<li>Three measurements of LV end systolic dimension, which appears as the shortest distance between the IVS and LVPW.</li>
			<li>Three measurements of heart rate, which is done by clicking the heart icon and measuring from systolic peak to systolic peak of the LVPW.</li>
			<li>If the RV lumen is dilated, obtain three measurements using the diagonal line icon.</li>
		</ol>
		</li>
		<li>Record a video loop of the short parasternal axis view in &#34;B mode&#34; by pressing the &#34;Cine Store&#34; button.</li>
		<li>Go to &#34;File&#34;, select &#34;Browse Study&#34; to recap your measurements, click &#34;End Session&#34;, and then &#34;Commit Session Data.&#34;</li>
		<li>Recover the mouse as outlined by the IACUC protocol, and clean up.</li>

Elevated pulmonary pressure and right ventricular (RV) dysfunction are the hallmarks of pulmonary vascular disease in animal models and human patients with pulmonary arterial hypertension (PAH). Transgenic and toxic (e.g. monocrotaline or hypoxia) models of PAH are widely used to study the pathophysiology of PAH and to investigate potential therapies. Given the expense and time involved in creating animal models of disease, it is critical that researchers have tools to accurately assess phenotypic expression of disease.
Echocardiography is the mainstay of the noninvasive assessment of ventricular function in rodent models1,2. Echocardiography has the advantage of clear translation to humans in whom the same tool is used. In addition, some genetic models exhibit incomplete penetrance3; the ability to noninvasively identify affected animals saves valuable time and resources. Noninvasive assessment of disease severity without sacrificing an animal also allows researchers to serially study the effects of investigative therapies. This is especially important given the rapidity with which translational therapies can progress to human trials4,5.
In humans, echocardiographic assessment of RV size and pulmonary hypertension is particularly challenging due to the retrosternal position and irregular shape of the RV6. Rodent models have the added challenges of small size and extremely rapid heart rates (300-700 beat/min). Recent advances including higher frame rates and smaller transducers have improved image quality and even allowed conscious imaging in some experimental protocols, though most rodent imaging is done under anesthesia7,8. Excellent experimental protocols of echocardiography in rat models of PAH have been described and validated against both MRI and invasive hemodynamics1,9. However, published echocardiography protocols in murine models of PAH are lacking.
In this article, we describe a protocol for assessing RV and pulmonary vascular function in a mouse model of PAH with a dominant negative BMPRII mutation and a model of isolated RV afterload after pulmonary artery banding; however, this protocol is applicable to any diseases affecting the pulmonary vasculature or right heart. We will describe animal preparation and detailed assessment of RV size and function as well as main pulmonary artery (PA) size. We also demonstrate the techniques and calculations needed to estimate stroke volume and cardiac output. Technical limitations preclude accurate Doppler estimates of pulmonary pressure, but we have applied a well-validated human surrogate, pulmonary artery acceleration time, to estimate PA pressure.

Transgenic and toxic models of pulmonary arterial hypertension (PAH) are widely used to study the pathophysiology of PAH and to investigate potential therapies. Given the expense and time involved in creating animal models of disease, it is critical that researchers have tools to accurately assess phenotypic expression of disease. Right ventricular dysfunction is the major manifestation of pulmonary hypertension. Echocardiography is the mainstay of the noninvasive assessment of right ventricular function in rodent models and has the advantage of clear translation to humans in whom the same tool is used. Published echocardiography protocols in murine models of PAH are lacking.
In this article, we describe a protocol for assessing RV and pulmonary vascular function in a mouse model of PAH with a dominant negative BMPRII mutation; however, this protocol is applicable to any diseases affecting the pulmonary vasculature or right heart. We provide a detailed description of animal preparation, image acquisition and hemodynamic calculation of stroke volume, cardiac output and an estimate of pulmonary artery pressure.

The principal goals of this protocol are to quantify RV size and function, and to understand the degree to which the pulmonary vasculature is diseased. Appropriate preparation of both the mouse and echocardiography equipment is essential to obtaining accurate and reproducible results. Magnetic Resonance Derived Myocardial Strain Assessment Using Feature Tracking

Purpose: An accurate and practical method to measure parameters like strain in myocardial tissue is of great clinical value, since it has been shown, that strain is a more sensitive and earlier marker for contractile dysfunction than the frequently used parameter EF. Current technologies for CMR are time consuming and difficult to implement in clinical practice. Feature tracking is a technology that can lead to more automization and robustness of quantitative analysis of medical images with less time consumption than comparable methods.
Methods: An automatic or manual input in a single phase serves as an initialization from which the system starts to track the displacement of individual patterns representing anatomical structures over time. The specialty of this method is that the images do not need to be manipulated in any way beforehand like e.g. tagging of CMR images.
Results: The method is very well suited for tracking muscular tissue and with this allowing quantitative elaboration of myocardium and also blood flow.
Conclusions: This new method offers a robust and time saving procedure to quantify myocardial tissue and blood with displacement, velocity and deformation parameters on regular sequences of CMR imaging. It therefore can be implemented in clinical practice.

Heterotopic Cervical Heart Transplantation in Mice

The heterotopic cervical heart transplantation in mice is a valuable tool in transplant and cardiovascular research. The cuff technique greatly simplifies this model by avoiding challenging suture anastomoses of small vessels thereby reducing warm ischemia time. In comparison to abdominal graft implantation the cervical model is less invasive and the implanted graft is easily accessible for further follow-up examinations. Anastomoses are performed by pulling the ascending aorta of the graft over the cuff with the recipient&#8217;s common carotid artery and by pulling the main pulmonary artery over the cuff with the external jugular vein. Selection of appropriate cuff size and complete mobilization of the vessels are important for successful revascularization. Ischemia-reperfusion (I/R) injury can be minimized by perfusing the graft with a cardioplegic solution and by hypothermia. In this article, we provide technical details for a simplified and improved cuff technique, which should allow surgeons with basic microsurgical skills to perform the procedure with a high success rate.

The cuff technique shortens and facilitates the model of heterotopic cervical heart transplantation in mice by avoiding technically challenging suture anastomoses of small vessels. Chronic Thromboembolic Pulmonary Hypertension and Assessment of Right Ventricular Function in the Piglet

An original piglet model of Chronic Thromboembolic Pulmonary Hypertension (CTEPH) associated with chronic Right Ventricular (RV) dysfunction is described. Pulmonary Hypertension (PH) was induced in 3-week-old piglets by a progressive obstruction of the pulmonary vascular bed. A ligation of the left Pulmonary Artery (PA) was performed first through a mini-thoracotomy. Second, weekly embolizations of the right lower pulmonary lobe were done under fluoroscopic guidance with n-butyl-2-cyanoacrylate during 5 weeks. Mean Pulmonary Arterial Pressure (mPAP) measured by ritght heart catheterism, increased progressively, as well as Right Atrial pressure and Pulmonary Vascular Resistances (PVR) after 5 weeks compared to sham animals. Right Ventricular (RV) structural and functional remodeling were assessed by transthoracic echocardiography (RV diameters, RV wall thickness, RV systolic function). RV elastance and RV-pulmonary coupling were assessed by Pressure-Volume Loops (PVL) analysis with conductance method. Histologic study of the lung and the right ventricle were also performed. Molecular analyses on RV fresh tissues could be performed through repeated transcutaneous endomyocardial biopsies. Pulmonary microvascular disease in obstructed and unobstructed territories was studied from lung biopsies using molecular analyses and pathology. Furthermore, the reliability and the reproducibility was associated with a range of PH severity in animals. Most aspects of the human CTEPH disease were reproduced in this model, which allows new perspectives for the understanding of the underlying mechanisms (mitochondria, inflammation) and new therapeutic approaches (targeted, cellular or gene therapies) of the overloaded right ventricle but also pulmonary microvascular disease.

Assessment of Cardiac Morphological and Functional Changes in Mouse Model of Transverse Aortic Constriction by Echocardiographic Imaging

Transverse aortic constriction (TAC) in mice has been used as a valuable model to study mechanisms of cardiac hypertrophy and heart failure1. A reliable noninvasive method is essential to assess real-time cardiac morphological and functional changes in animal models of heart disease. Transthoracic echocardiography represents an important tool for noninvasive assessment of cardiac structure and function2. Here we used a high-resolution ultrasound imaging system to monitor myocardial remodeling and heart failure progression over time in a mouse model of TAC. B-mode, M-mode, and Doppler imaging were used to precisely assess cardiac hypertrophy, ventricular dilatation, and functional deterioration in mice following TAC. Color and pulse wave (PW) Doppler imaging was used to noninvasively measure pressure gradient across the aortic constriction created by TAC and to assess transmitral blood flow in mice. Thus transthoracic echocardiographic imaging provides comprehensive noninvasive measurements of cardiac dimensions and function in mouse models of heart disease.

Echocardiographic Approaches and Protocols for Comprehensive Phenotypic Characterization of Valvular Heart Disease in Mice

The aim of this manuscript and accompanying video is to provide an overview of the methods and approaches used for imaging heart valve function in rodents, with detailed descriptions of the appropriate methods for anesthesia, the echocardiographic windows used, the imaging planes and probe orientations for image acquisition, the methods for data analysis, and the limitations of emerging technologies for the evaluation of cardiac and valvular function. Importantly, we also highlight several future areas of research in cardiac and heart valve imaging that may be leveraged to gain insights into the pathogenesis of valve disease in preclinical animal models. We propose that using a systematic approach to evaluating cardiac and heart valve function in mice can result in more robust and reproducible data, as well as facilitate the discovery of previously underappreciated phenotypes in genetically-altered and/or physiologically-stressed mice.

Echocardiographic and Histological Examination of Cardiac Morphology in the Mouse

An increasing number of genetically modified mouse models has become available in recent years. Moreover, the number of pharmacological studies performed in mice is high. Phenotypic characterization of these mouse models also requires the examination of cardiac function and morphology. Echocardiography and magnetic resonance imaging (MRI) are commonly used approaches to characterize cardiac function and morphology in mice. Echocardiographic and MRI equipment specialized for use in small rodents is&#160;expensive and requires a dedicated space. This protocol describes cardiac measurements in mice using a clinical echocardiographic system with a 15 MHz human vascular probe. Measurements are performed on anesthetized adult mice. At least three image sequences are recorded and analyzed for each animal in M-mode in the parasternal short-axis view. Afterwards, cardiac histological examination is performed, and cardiomyocyte diameters are determined on hematoxylin-eosin- or wheat germ agglutinin (WGA)-stained paraffin sections. Vessel density is determined morphometrically after Pecam-1 immunostaining. The protocol has been applied successfully to pharmacological studies and different genetic animal models under baseline conditions, as well as after experimental myocardial infarction by the permanent ligation of the left anterior descending coronary artery (LAD). In our experience, echocardiographic investigation is limited to anesthetized animals and is feasible in adult mice weighing at least 25 g.

Echocardiographic Measurement of Right Ventricular Diastolic Parameters in Mouse

Diastolic dysfunction is a prominent feature of right ventricular (RV) remodeling associated with conditions of pressure overload. However, the RV diastolic function is rarely quantified in experimental studies. This might be due to technical difficulties in the visualization of the RV in the apical four-chamber view in rodents. Here we describe two positions facilitating the visualization of the apical four-chamber view in mice to assess the RV diastolic function.
The apical four-chamber view is enabled by tilting the mouse fixation platform to the left and caudally (LeCa) or to the right and cranially (RiCr). Both positions provide images of comparable quality. The results of the RV diastolic function obtained from two positions are not significantly different. Both positions are comparably easy to perform. This protocol can be incorporated into published protocols and enables detailed investigations of the RV function.

Assessment and Characterization of Hyaloid Vessels in Mice

In the eye, the embryonic hyaloid vessels nourish the developing lens and retina and regress when the retinal vessels develop. Persistent or failed regression of hyaloid vessels can be seen in diseases such as persistent hyperplastic primary vitreous (PHPV), leading to an obstructed light path and impaired visual function. Understanding the mechanisms underlying the hyaloid vessel regression may lead to new molecular insights into the vascular regression process and potential new ways to manage diseases with persistent hyaloid vessels. Here we describe the procedures for imaging hyaloid in live mice with optical coherence tomography (OCT) and fundus fluorescein angiography (FFA) and a detailed technical protocol of isolating and flat-mounting hyaloid ex vivo for quantitative analysis. Low-density lipoprotein receptor-related protein 5 (LRP5) knockout mice were used as an experimental model of persistent hyaloid vessels, to illustrate the techniques. Together, these techniques may facilitate a thorough assessment of hyaloid vessels as an experimental model of vascular regression and studies on the mechanism of persistent hyaloid vessels.

Transthoracic Echocardiographic Examination in the Rabbit Model

Large animal models such as the rabbit are valuable for translational preclinical research. Rabbits have a similar cardiac electrophysiology compared to that of humans and that of other large animal models such as dogs and pigs. However, the rabbit model has the additional advantage of lower maintenance costs compared to other large animal models. The longitudinal evaluation of cardiac function using echocardiography, when appropriately implemented, is a useful methodology for preclinical assessment of novel therapies for heart failure with reduced ejection fraction (e.g. cardiac regeneration). The correct use of this non-invasive tool requires the implementation of a standardized examination protocol following international guidelines. Here we describe, step by step, a detailed protocol supervised by veterinary cardiologists for performing echocardiography in the rabbit model, and demonstrate how to correctly obtain the different echocardiographic views and imaging planes, as well as the different imaging modes available in a clinical echocardiography system routinely used in human and veterinary patients.

Invasive Hemodynamic Assessment for the Right Ventricular System and Hypoxia-Induced Pulmonary Arterial Hypertension in Mice

Pulmonary arterial hypertension (PAH) is a chronic and severe cardiopulmonary disorder. Mice are a popular animal model used to mimic this disease. However, the evaluation of right ventricular pressure (RVP) and pulmonary artery pressure (PAP) remains technically challenging in mice. RVP and PAP are more difficult to measure than left ventricular pressure because of the anatomical differences between the left and right heart systems. In this paper, we describe a stable right heart hemodynamic measurement method and its validation using healthy and PAH mice. This method is based on open-chest surgery and mechanical ventilation support. It is a complicated procedure compared to closed chest procedures. While a well-trained surgeon is required for this surgery, the advantage of this procedure is that it can generate both RVP and PAP parameters at the same time, so it is a preferable procedure for the evaluation of PAH models.
