1.&#160;Preparation and Calculations
<ol>
	<li>Prepare 1 mM HCl solution using concentrated hydrochloric acid, 37%, and water. 
	Note: Caution should always be used when handling concentrated acids. Concentrated acid should always be added to water, never add water to concentrated acid.</li>
	<li>Determine the desired concentration of TMOS, siloxane precursor, for the microwave reaction. A TMOS concentration of 25 mM will be used for procedural demonstrations.</li>
</ol>
2.&#160;Hydrolysis of TMOS and Preparation of Reaction Solution
<ol>
	<li>For a 25 mM reaction solution of TMOS in acetone, obtain a 50 ml&#160;plastic conical tube, TMOS, and the prepared 1 mM HCl solution.</li>
	<li>Using a micropipetter, add 850 &#181;l&#160;of the 1 mM HCl solution to the plastic conical tube.</li>
	<li>Using a micropipetter, add 150 &#181;l&#160;of TMOS to the plastic conical tube containing the 850 &#181;l&#160;of 1 mM HCl.</li>
	<li>Observe that the mixture of TMOS and acid are originally immiscible, resulting in two clear colorless layers.</li>
	<li>Vortex the solution. A quick burst will cause solution to turn slightly cloudy and pale white. Vortexing the solution again will yield a clear colorless solution. Total vortex time is ~10 sec. At this point, the TMOS is hydrolyzed and in the form of silicic acid.</li>
	<li>Add acetone to the plastic conical tube containing the silicic acid solution and dilute to a total volume of 40 ml. Ensure proper mixing by vortexing the solution after diluting the solution with acetone.</li>
</ol>
Note: Prior to microwave heating, the silicic acid / acetone solution will be stable for several hours. Silica condensation was not observed in similar prepared solutions which were stable for several weeks without any evidence of condensation. Although for best results, reaction solutions should be used upon day of preparation to ensure reproducible results.
3.&#160;Create Microwave Method on Microwave Reactor
<ol>
	<li>Create a microwave method within the provided software for SiO2 NP formation. Typical reaction parameters include: power = 300 W, temperature = 125 &#176;C and time = 60 sec. Using an external laptop the temperature, pressure, and power profiles can be stored, plotted, and compared with future experiments as in Figure 1.</li>
</ol>
4.&#160;Microwave-assisted Synthesis of SiO2&#160;Nanoparticles
<ol>
	<li>Obtain a 10 ml&#160;microwave reaction vial, stir bar, and snap cap.</li>
	<li>Add the stir bar to the vial and using a micropipetter add 5 ml&#160;of the silicic acid/acetone reaction solution to the microwave vial. A reaction volume of 5 ml&#160;should be used to provide adequate headspace to accommodate the generation of pressure from superheated acetone.</li>
	<li>Place the snap cap on 10 ml&#160;vial. Ensure cap seats properly on the vial to reduce loss of solvent when superheated.</li>
	<li>Place vial in microwave reactor.</li>
	<li>Press the start button on the laptop to start the microwave heating procedure. Observe proper placement of pressure head unit. Observe that the proper microwave power settings are correct once the reaction begins: correct power setting, increase in reaction temperature, and generation of reaction pressure. When the reaction time has expired, compressed air will quickly cool the reaction.</li>
	<li>Remove the reaction vial from microwave reactor after cooling.</li>
</ol>
Note: Typically for a 5 ml&#160;reaction, a measured conversion of ~40-50% has been reported19 in the preparation of SiO2 NPs by these microwave-assisted methods.
5.&#160;DLS Size Analysis of Reaction Solutions
<ol>
	<li>Obtain a polystyrene disposable cuvette.</li>
	<li>Add 900 &#181;l&#160;of distilled water to the cuvette.</li>
	<li>Add 100 &#181;l&#160;of reaction solution containing SiO2 NP to the cuvette containing the water and mix.</li>
	<li>Setup procedures for particle measurements by following DLS analyzer software and analyze the solution for particle size.</li>
</ol>
6.&#160;SiO2 NP Cleanup Procedure
<ol>
	<li>Add 1 ml&#160;of SiO2 NP reaction solution to an Eppendorf tube.</li>
	<li>Centrifuge at 15.7k x g for 30-60 min&#160;depending on particle diameter.</li>
	<li>Carefully decant excess solution from SiO2 NP pellet.</li>
	<li>Add 1 ml&#160;fresh acetone and resuspend SiO2 NP by placing in an ultrasonic bath for 5-10 min.</li>
	<li>Repeat (centrifuging, decanting, resuspending) two more times for adequate removal of residual silicic acid.</li>
</ol>
7.&#160;SEM Sample Preparation
<ol>
	<li>Clean the highly polished single crystal silicon wafers in an ultrasonic bath for approximately 30 min prior to use.</li>
	<li>Place the silicon wafers in piranha solution (3:1 conc. H2SO4:30% H2O2) at a temperature of 80 &#176;C for 1 hr after removing from the ultrasonic bath. Wash the silicon wafers with distilled water and dry. 
	Note: Caution should be used when handling concentrated acids, especially piranha solution.</li>
	<li>Drop-cast clean or not clean solutions of SiO2 NPs onto the clean prepared silicon wafer.</li>
	<li>Place prepared samples on silicon wafer in the cylindrical tube of the sputter system.</li>
	<li>Vacuum the cylindrical tube of the sputter system to 40 mTorr.</li>
	<li>Purge the cylindrical tube of the sputter system with Argon gas until a pressure of approximately 200 mTorr is reached.</li>
	<li>Decrease the pressure to 80 mTorr and apply 15 mA of voltage and keep constant. Sputter platinum on prepared samples for 60 sec.</li>

The microwave-assisted methods described in this manuscript are advantageous over conventional heating methods because SiO2 NPs can be synthesized accurately, precisely, and quickly. The following criteria should be followed to eliminate any potential issues associated with SiO2 NPs formation by these microwave-associated techniques: 1) use of a catalyst such as 1 mM HCl, 2) hydrolysis of the TMOS should be completed before addition of acetone, 3) use of an aprotic solvent such as acetone, 4) use of...

Silica nanoparticles (SiO2 NPs) were first synthesized by St&#246;ber1 and through modifications2-7 have become the preferred method for SiO2 NPs synthesis. Typically, St&#246;ber reactions are catalyzed by alkaline conditions where silica sols are formed. Acid-catalyzed reactions are used less frequently than alkaline-catalyzed reactions due to the greater degree of difficulty of hydrolysis of the siloxane precursor. Unlike alkaline-catalyzed reactions, acid-catalyzed reactions preferentially form silica gels.8
Microwave-assisted chemical reactions are an emerging technique within the scientific community and in literature due to the associated benefits to the techniques9-18. Specifically, microwave-assisted techniques have been shown to be advantageous in the synthesis of nanomaterials where the promotion of spontaneous nucleation events is desired. Microwave conditions are advantageous because microwave reactors deliver controlled power quickly to the reaction10. Until recently19, the synthesis of SiO2 NPs using microwave reactors have been used with limited success mainly as a result of issues with reproducibility20-22.
The details and procedural methods often reported in the literature on the synthesis of nanomaterials often tend to be obscure and sometimes seen as an &#34;art-form.&#34; Combining microwave-assisted synthetic techniques and nanomaterial synthesis can compound the subject even further. The purpose of this manuscript is to guide researchers in the synthesis of nanomaterials by microwave-assisted techniques, eliminate obscurity associated with these techniques, and point out common mistakes associated with these techniques.
In a microwave chemical reaction, any molecular species containing a permanent dipole is capable of interacting and perturbing the electromagnetic (EM) field. These species are not limited solely to the reagents and solvents used within the reaction, but can be any substance placed in the EM field i.e. glass vials, salts, ionic liquids.
The ability for a specific substance to effectively convert EM energy into heat is defined as the loss factor of a material or tan &#948;. Solvents are commonly classified by their loss factor where values for tan &#948; &#62; 0.5 are considered high, 0.1 &#62; tan &#948; &#62; 0.5 are considered medium, and tan &#948; &#60; 0.1 are considered low. These loss factors values relate the ability for a particular solvent to couple or absorb microwave energy and convert that energy into heat. Thermal energy is generated through molecular friction of species attempting to align with the oscillating EM field. If solvents with high tan &#948; values are used within a reaction, the solvent will dominate the microwave absorption events, masking the precursor or reagents, leading to bulk heating as a result of the solvent strongly coupling with the EM field.
Typically, polar solvents are used in SiO2 NP synthesis to ensure reagent solubility and for donation of labile protons23. Common solvents used in SiO2 NP syntheses are alcohol solvents such as ethanol, methanol or 2-propanol. These solvents all have high tan &#948; values (0.941, 0.799, and 0.659 for ethanol, 2-propanol and methanol, respectively) making them poor solvent choices for microwave-assisted chemical reactions of SiO2 NPs as they efficiently couple with the EM field. It is our belief that microwave-assisted reactions are most effective when low tan &#948; solvents are used in combination with polar molecular precursors in synthetic reactions. These circumstances allow for the molecular precursors to couple with the EM field, providing molecular heating, while the solvent interacts minimally. For microwave-assisted reactions in this manuscript, acetone is used as an alternative to the commonly used alcohol solvents associated with SiO2 NPs synthesis. Acetone is considered a low loss factor solvent (tan &#948; = 0.054), which limits the solvent interactions within the EM field allowing selective microwave absorption with the reactants meditating silica condensation reactions.
In this manuscript, we outline the procedures associated with the microwave-assisted synthesis of SiO2 NPs which are accurate, precise and quick. SiO2 NP growth is achieved by effective coupling of the precursor with the EM field while the solvent has a minimal role in heating. Hydrolysis of the siloxane precursor is achieved by using hydrochloric acid, which leads to slower rates of hydrolysis and limits further condensation reactions. Alkaline-catalyzed reactions have much faster reaction rates and can complicate growth processes when used with microwave-assisted techniques. The resultant SiO2 NPs synthesized by these techniques range in diameters from 30 nm up to diameters greater than 250 nm. SiO2 NP size is controlled by varying the precursor concentration and exposure time to the microwave radiation.

Microwave-assisted synthetic techniques were used to quickly and reproducibly produce silica nanoparticle sols using an acid catalyst with nanoparticle diameters ranging from 30-250 nm by varying the reaction conditions. Through the selection of a microwave compatible solvent, silicic acid precursor, catalyst, and microwave irradiation time, these microwave-assisted methods were capable of overcoming the previously reported shortcomings associated with synthesis of silica nanoparticles using microwave reactors. The siloxane precursor was hydrolyzed using the acid catalyst, HCl. Acetone, a low-tan &#948; solvent, mediates the condensation reactions and has minimal interaction with the electromagnetic field. Condensation reactions begin when the silicic acid precursor couples with the microwave radiation, leading to silica nanoparticle sol formation. The silica nanoparticles were characterized by dynamic light scattering data and scanning electron microscopy, which show the materials&#39; morphology and size to be dependent on the reaction conditions. Microwave-assisted reactions produce silica nanoparticles with roughened textured surfaces that are atypical for silica sols produced by St&#246;ber&#39;s methods, which have smooth surfaces.

Temperature, pressure, and microwave power traces for a representative SiO2 NP microwave-assisted reaction are presented in Figure 1. Microwave-assisted Intramolecular Dehydrogenative Diels-Alder Reactions for the Synthesis of Functionalized Naphthalenes/Solvatochromic Dyes

Functionalized naphthalenes have applications in a variety of research fields ranging from the synthesis of natural or biologically active molecules to the preparation of new organic dyes. Although numerous strategies have been reported to access naphthalene scaffolds, many procedures still present limitations in terms of incorporating functionality, which in turn narrows the range of available substrates. The development of versatile methods for direct access to substituted naphthalenes is therefore highly desirable.
The Diels-Alder (DA) cycloaddition reaction is a powerful and attractive method for the formation of saturated and unsaturated ring systems from readily available starting materials. A new microwave-assisted intramolecular dehydrogenative DA reaction of styrenyl derivatives described herein generates a variety of functionalized cyclopenta[b]naphthalenes that could not be prepared using existing synthetic methods. When compared to conventional heating, microwave irradiation accelerates reaction rates, enhances yields, and limits the formation of undesired byproducts.
The utility of this protocol is further demonstrated by the conversion of a DA cycloadduct into a novel solvatochromic fluorescent dye via a Buchwald-Hartwig palladium-catalyzed cross-coupling reaction. Fluorescence spectroscopy, as an informative and sensitive analytical technique, plays a key role in research fields including environmental science, medicine, pharmacology, and cellular biology. Access to a variety of new organic fluorophores provided by the microwave-assisted dehydrogenative DA reaction allows for further advancement in these fields.

Microwave-assisted Functionalization of Poly(ethylene glycol) and On-resin Peptides for Use in Chain Polymerizations and Hydrogel Formation

One of the main benefits to using poly(ethylene glycol) (PEG) macromers in hydrogel formation is synthetic versatility. The ability to draw from a large variety of PEG molecular weights and configurations (arm number, arm length, and branching pattern) affords researchers tight control over resulting hydrogel structures and properties, including Young&#8217;s modulus and mesh size. This video will illustrate a rapid, efficient, solvent-free, microwave-assisted method to methacrylate PEG precursors into poly(ethylene glycol) dimethacrylate (PEGDM). This synthetic method provides much-needed starting materials for applications in drug delivery and regenerative medicine. The demonstrated method is superior to traditional methacrylation methods as it is significantly faster and simpler, as well as more economical and environmentally friendly, using smaller amounts of reagents and solvents. We will also demonstrate an adaptation of this technique for on-resin methacrylamide functionalization of peptides. This on-resin method allows the N-terminus of peptides to be functionalized with methacrylamide groups prior to deprotection and cleavage from resin. This allows for selective addition of methacrylamide groups to the N-termini of the peptides while amino acids with reactive side groups (e.g.&#160;primary amine of lysine, primary alcohol of serine, secondary alcohols of threonine, and phenol of tyrosine) remain protected, preventing functionalization at multiple sites. This article will detail common analytical methods (proton Nuclear Magnetic Resonance spectroscopy (;H-NMR) and Matrix Assisted Laser Desorption Ionization Time of Flight mass spectrometry (MALDI-ToF)) to assess the efficiency of the functionalizations. Common pitfalls and suggested troubleshooting methods will be addressed, as will modifications of the technique which can be used to further tune macromer functionality and resulting hydrogel physical and chemical properties. Use of synthesized products for the formation of hydrogels for drug delivery and cell-material interaction studies will be demonstrated, with particular attention paid to modifying hydrogel composition to affect mesh size, controlling hydrogel stiffness and drug release.

One-pot Microwave-assisted Conversion of Anomeric Nitrate-esters to Trichloroacetimidates

The goal of the following procedure is to provide a demonstration of the one-pot conversion of a 2-azido-1-nitrate-ester to a trichloroacetimidate glycosyl donor. Following azido-nitration of a glycal, the product 2-azido-1-nitrate ester can be hydrolyzed under microwave-assisted irradiation. This transformation is usually achieved using strongly nucleophilic reagents and extended reaction times. Microwave irradiation induces hydrolysis, in the absence of reagents, with short reaction times. Following denitration, the intermediate anomeric alcohol is converted, in the same pot, to the corresponding 2-azido-1-trichloroacetimidate.

A 2-azido-1-nitrate-ester can be converted to the corresponding 2-azido-1-trichloroacetimidate in a one-pot procedure. The goal of the manuscript is to demonstrate utility of the microwave reactor in carbohydrate synthesis.

Preparation of Functional Silica Using a Bioinspired Method

The goal of the protocols described herein is to synthesize bioinspired silica materials, perform enzyme encapsulation therein, and partially or totally purify the same by acid elution. By combining sodium silicate with a polyfunctional bioinspired additive, silica is rapidly formed at ambient conditions upon neutralization. 
The effect of neutralization rate and biomolecule addition point on silica yield are investigated, and biomolecule immobilization efficiency is reported for varying addition point. In contrast to other porous silica synthesis methods, it is shown that the mild conditions required for bioinspired silica synthesis are fully compatible with the encapsulation of delicate biomolecules. Additionally, mild conditions are used across all synthesis and modification steps, making bioinspired silica a promising target for the scale-up and commercialization as both a bare material and active support medium.
The synthesis is shown to be highly sensitive to conditions, i.e., the neutralization rate and final synthesis pH, however tight control over these parameters is demonstrated through the use of auto titration methods, leading to high reproducibility in reaction progression pathway and yield. 
Therefore, bioinspired silica is an excellent active material support choice, showing versatility towards many current applications, not limited to those demonstrated here, and potency in future applications.

Here, we present a protocol to synthesize bioinspired silica materials and immobilize enzymes therein. Silica is synthesized by combining sodium silicate and an amine &#39;additive&#39;, which neutralize at a controlled rate. Material properties and function can be altered either by in situ enzyme immobilization or post-synthetic acid elution of encapsulated additives.

Reducing Willow Wood Fuel Emission by Low Temperature Microwave Assisted Hydrothermal Carbonization

Biomass is a sustainable fuel, as its CO2 emissions are reintegrated in biomass growth. However, the inorganic precursors in the biomass cause a negative environmental impact and slag formation. The selected short rotation coppice (SRC) willow wood has a high ash content (<img alt="Equation 1" src="/files/ftp_upload/58970/58970eq1.jpg" /> = 1.96%) and, therefore, a high content of emission and slag precursors. Therefore, the reduction of minerals from SRC willow wood by low temperature microwave assisted hydrothermal carbonization (MAHC) at 150 &#176;C, 170 &#176;C, and 185 &#176;C is investigated. An advantage of MAHC over conventional reactors is an even temperature conductance in the reaction medium, as microwaves penetrate the whole reactor volume. This allows a better temperature control and a faster cooldown. Therefore, a succession of depolymerization, transformation and repolymerization reactions can be analyzed effectively. In this study, the analysis of the mass loss, ash content and composition, heating values and molar O/C and H/C ratios of the treated and untreated SCR willow wood showed that the mineral content of the MAHC coal was reduced and the heating value increased. The process water showed a decreasing pH and contained furfural and 5-methylfurfural. A process temperature of 170 &#176;C showed the best combination of energy input and ash component reduction. The MAHC allows a better understanding of the hydrothermal carbonization process, while a large-scale industrial application is unlikely because of the high investment costs.

Microwave-Assisted Preparation of 1-Aryl-1H-pyrazole-5-amines

A synthetic process for the preparation of a variety of 1-aryl-1H-pyrazole-5-amines was developed. The microwave-mediated nature of this method makes it efficient in both time and resources and utilizes water as the solvent. 3-Aminocrotononitrile or an appropriate &#945;-cyanoketone is combined with an aryl hydrazine and dissolved in 1 M HCl. The mixture is then heated in a microwave reactor at 150 &#176;C, typically for 10-15 min. The product can be readily obtained by basifying the solution with 10% NaOH and isolating the desired compound with a simple vacuum filtration. The use of water as a solvent in this reaction lends to its ease and utility in production, and this method is easily reproducible with a variety of functional groups. Typical isolated yields range from 70-90%, and reactions can be performed on the milligram to gram scale with little to no change in observed yields. Some of the applications of these molecules and their derivatives include pesticides, anti-malarials, and chemotherapeutics, among many others.

Microwave-Assisted Preparation of 1-Aryl-1H-pyrazole-5-amines

1-Aryl-1H-pyrazole-5-amines are prepared from aryl hydrazines combined with either 3-aminocrotononitrile or an &#945;-cyanoketone in a 1 M HCl solution using a microwave reactor. Most reactions are done in 10-15 minutes and pure product can be obtained via vacuum filtration with typical isolated yields of 70-90%.

A Microwave-Assisted Direct Heteroarylation of Ketones Using Transition Metal Catalysis

Heteroarylation introduces heteroaryl fragments to organic molecules. Despite the numerous available reactions reported for arylation via transition metal catalysis, the literature on direct heteroarylation is scarce. The presence of heteroatoms such as nitrogen, sulfur and oxygen often make heteroarylation a challenging research field due to catalyst poisoning, product decomposition and the rest. This protocol details a highly efficient direct &#945;-C(sp3) heteroarylation of ketones under microwave irradiation. Key factors for successful heteroarylation include the use of XPhos Palladacycle Gen. 4 Catalyst, excess base to suppress side reactions and the high temperature and pressure achieved in a sealed reaction vial under microwave irradiation. The heteroarylation compounds prepared by this method were fully characterized by proton nuclear magnetic resonance spectroscopy (1H NMR), carbon nuclear magnetic resonance spectroscopy (13C NMR) and high-resolution mass spectrometry (HRMS). This methodology has several advantages over literature precedents including broad substrate scope, rapid reaction time, greener procedure and operational simplicity by eliminating the preparation of intermediates such as silyl enol ether. Possible applications for this protocol include, but are not limited to, diversity-oriented synthesis for the discovery of biologically active small molecules, domino synthesis for the preparation of natural products and ligand development for new transition metal catalytic systems.

Preparation of Nanoparticles for ToF-SIMS and XPS Analysis

Nanoparticles have gained increasing attention in recent years due to their potential and application in different fields including medicine, cosmetics, chemistry, and their potential to enable advanced materials. To effectively understand and regulate the physico-chemical properties and potential adverse effects of nanoparticles, validated measurement procedures for the various properties of nanoparticles need to be developed. While procedures for measuring nanoparticle size and size distribution are already established, standardized methods for analysis of their surface chemistry are not yet in place, although the influence of the surface chemistry on nanoparticle properties is undisputed. In particular, storage and preparation of nanoparticles for surface analysis strongly influences the analytical results from various methods, and in order to obtain consistent results, sample preparation must be both optimized and standardized. In this contribution, we present, in detail, some standard procedures for preparing nanoparticles for surface analytics. In principle, nanoparticles can be deposited on a suitable substrate from suspension or as a powder. Silicon (Si) wafers are commonly used as substrate, however, their cleaning is critical to the process. For sample preparation from suspension, we will discuss drop-casting and spin-coating, where not only the cleanliness of the substrate and purity of the suspension but also its concentration play important roles for the success of the preparation methodology. For nanoparticles with sensitive ligand shells or coatings, deposition as powders is more suitable, although this method requires particular care in fixing the sample.

A number of different procedures for preparing nanoparticles for surface analysis are presented (drop casting, spin coating, deposition from powders, and cryofixation). We discuss the challenges, opportunities, and possible applications of each method, particularly regarding the changes in the surface properties caused by the different preparation methods.

Microwave Synthesis and Characterization of Nickel Hydroxide Nanosheets

Effect of Microwave Synthesis Conditions on the Structure of Nickel Hydroxide Nanosheets

A protocol for rapid, microwave-assisted hydrothermal synthesis of nickel hydroxide nanosheets under mildly acidic conditions is presented, and the effect of reaction temperature and time on the material&#39;s structure is examined. All reaction conditions studied result in aggregates of layered &#945;-Ni(OH)2 nanosheets. The reaction temperature and time strongly influence the structure of the material and product yield. Synthesizing &#945;-Ni(OH)2 at higher temperatures increases the reaction yield, lowers the interlayer spacing, increases crystalline domain size, shifts the frequencies of interlayer anion vibrational modes, and lowers the pore diameter. Longer reaction times increase reaction yields and result in similar crystalline domain sizes. Monitoring the reaction pressure in situ shows that higher pressures are obtained at higher reaction temperatures. This microwave-assisted synthesis route provides a rapid, high-throughput, scalable process that can be applied to the synthesis and production of a variety of transition metal hydroxides used for numerous energy storage, catalysis, sensor, and other applications.

Following homogenization, the sample undergoes acid digestion, wherein it is digested with acids and auxiliary chemicals at elevated temperatures, transforming solid samples into a liquid state. In this process, metals in the original sample react with acids to form water-soluble salts. Samples prepared through acid digestion are suitable for elemental analysis using techniques such as inductively coupled plasma mass spectrometry, inductively coupled plasma optical emission spectroscopy, atomic absorption spectroscopy, electrochemical methods, and other analytical techniques. This work details the preparation of food samples for multi-element determination using inductively coupled plasma mass spectrometry. The step-by-step procedure involves the homogenization process using a laboratory-sized mixer with ceramic blades, followed by acid digestion in closed vessels using microwave-assisted wet acid digestion.
