&#x6811;&#x7A81;&#x72B6;&#x91CA;&#x653E;&#x591A;&#x5DF4;&#x80FA;&#x7684;&#x4EE3;&#x8C22;&#x578B;&#x8C37;&#x6C28;&#x9178;&#x53D7;&#x4F53;&#x5728;&#x5927;&#x9F20;&#x8111;&#x5185;&#x9ED1;&#x8D28;&#x4E2D;&#x7684;&#x8C03;&#x8282;&#x4F5C;&#x7528;&#x3002;

A superfusion system was used to study the effects of metabotropic glutamate receptor (mGluR) ligands upon the release of [(3)H]dopamine ([(3)H]DA) previously taken up by rat substantia nigra (SN) slices. trans-(+/-)-1-Amino-(1S,3R)-cyclopentane dicarboxylic acid (trans-ACPD; 100 and 600 microM), a group I and II mGluR agonist, evoked the release of [(3)H]DA from nigral slices. This last effect was reduced significantly by (2S,3S,4S)-2-methyl-2-(carboxycyclopropyl)-glycine (MCCG; 300 microM), an antagonist of group II mGluR, or by the addition of tetrodotoxin (D-APV; 1 microM) to the superfusion medium. D-(-)-2-Amino-5-phosphono-valeric acid (100 microM), an N-methyl-D-aspartate receptor antagonist, or the presence of Mg(2+) (1.2mM) in the superfusion medium did not modify trans-ACPD-induced [(3)H]DA release. In addition, a group II mGluR agonist such as (2S,1'R,2'R,3'R)-2-(2',3'-dicarboxycyclopropyl)-glycine (DCG-IV; 100 microM) significantly induced the release of [(3)H]DA from nigral slices, whereas a group I mGluR agonist such as (RS)-3,5-dihydroxyphenylglycine (DHPG; 50 and 100 microM) did not modify the release of the [(3)H]-amine. Further experiments showed that the NMDA (100 microM)-evoked release of [(3)H]DA was decreased significantly by prior exposure of SN slices to trans-ACPD. Finally, partial denervation of the DA nigro-striatal pathway with 6-hydroxydopamine (6-OH-DA) increased trans-ACPD-induced release of [(3)H]DA, whereas it decreased trans-ACPD inhibitory effects on NMDA-evoked release of [(3)H]DA from nigral slices. The present results suggest that the dendritic release of DA in the SN is regulated by mGluR activation. Such nigral mGluR activation may produce opposite effects upon basal and NMDA-evoked release of DA in the SN. In addition, such mGluR-induced effects in the SN are modified in response to partial denervation of the DA nigro-striatal pathway.

Un syst&egrave;me de perfusion a &eacute;t&eacute; utilis&eacute; pour &eacute;tudier les effets des ligands des r&eacute;cepteurs (mGluR) glutamate m&eacute;tabotropique sur la lib&eacute;ration de [3 H] dopamine ([3 H] DA) pr&eacute;c&eacute;demment occup&eacute;s par tranches de substantia nigra (SN) de rat. trans-(+/-)-1-amino-(1s,3R)-cyclopentane acide dicarboxylique (trans-ACDP ; 100 et 600 microM), un groupe que j&#39;ai et agoniste de mGluR II, &eacute;voqu&eacute;e par la lib&eacute;ration de [3 H] DA des tranches de cortico. Ce dernier effet a &eacute;t&eacute; r&eacute;duit de fa&ccedil;on significative par (2S,3S,4S)-2-methyl-2-(carboxycyclopropyl)-glycine (MCCG ; 300 microM), un antagoniste du groupe II mGluR, ou par l&#39;addition de la t&eacute;trodotoxine (D-APV; 1 microM) au milieu de perfusion. Acide D-(-)-2-amino-5-phosphono-valeric (100 microM), un antagoniste des r&eacute;cepteurs N-m&eacute;thyl-D-aspartate ou la pr&eacute;sence de Mg(2+) (1,2 mM) dans le milieu de superfusion n&#39;a pas modifi&eacute; la lib&eacute;ration de [3 H] DA induite par trans-ACPD. En outre, un agoniste de mGluR groupe II comme le (2 s, 1&#39;R, R 2&#39;, 3&#39;R)-2-(2&#39;,3&#39;-dicarboxycyclopropyl)-glycine (DCG-IV; 100 microM) significativement induit la lib&eacute;ration de [3 H] DA des tranches de cortico, un groupe j&#39;ai mGluR agoniste comme (RS) - 3, 5 - dihydroxyphenylglycine (DHPG ; microM 50 et 100) ne modifia pas la lib&eacute;ration de la [3 H]-amine. D&#39;autres exp&eacute;riences ont montr&eacute; que le NMDA (100 microM)-lib&eacute;ration &eacute;voqu&eacute;e de [3 H] DA a diminu&eacute; consid&eacute;rablement par une exposition ant&eacute;rieure des tranches SN &agrave; trans-ACPD. Enfin, la d&eacute;nervation partielle de la voie nigro-striatale de DA &agrave; la 6-hydroxydopamine (6-OH-DA) augment&eacute; trans ACPD-lib&eacute;ration de [3 H] DA, alors qu&#39;elle diminue l&#39;action inhibitrice sur la lib&eacute;ration &eacute;voqu&eacute;e par NMDA de [3 H] DA des tranches de cortico trans-ACPD. Ces r&eacute;sultats sugg&egrave;rent que la lib&eacute;ration dendritique de DA dans le SN est r&eacute;gul&eacute;e par l&#39;activation mGluR. Cette activation de mGluR cortico peut produire des effets sur la lib&eacute;ration basale et &eacute;voqu&eacute;e par le NMDA de DA dans le SN oppos&eacute;s. En outre, ces effets dans le SN mGluR sont modifi&eacute;es en r&eacute;ponse &agrave; une d&eacute;nervation partielle de la voie nigro-striatale de DA.

Un sistema di Sopraffusione &egrave; stato utilizzato per studiare gli effetti di metabotropic glutammato ligandi del recettore (mGluR) subito dopo il rilascio di dopamina [3 H] ([(3) H] DA) precedentemente presi dalle fette di ratto substantia nigra (SN). Trans-(+/-)-1-amino-(1s,3R)-cyclopentane acido dicarbossilico (trans-ACPD; 100 e 600 microM), un gruppo I e agonista mGluR II, evocato il rilascio di [(3) H] DA fette nigral. Questo ultimo effetto &egrave; stato ridotto in modo significativo a (2S,3S,4S)-2-methyl-2-(carboxycyclopropyl)-glycine (MCCG; 300 microM), un antagonista del gruppo II mGluR, o con l&#39;aggiunta di tetrodotossina (D-APV; 1 microM) per il supporto di Sopraffusione. Acido D-(-)-2-amino-5-phosphono-Valeric (100 microM), un antagonista del recettore N-metil-D-aspartato o la presenza di Mg(2+) (1.2 mM) nel mezzo di Sopraffusione non ha fatto modificare rilascio [3 H] DA trans-ACPD-indotta. In aggiunta, un agonista di gruppo II mGluR come (2S, 1&#39;R, 2&#39;R, 3&#39;R)-2-(2&#39;,3&#39;-dicarboxycyclopropyl)-glicina (DCG-IV; 100 microM) indotta significativamente il rilascio di [(3) H] DA nigral fette, mentre un gruppo I mGluR agonista come (RS) - 3,5 - dihydroxyphenylglycine (DHPG; 50 e 100 microM) non ha fatto modificare il rilascio della (3) [H]-ammina. Ulteriori esperimenti hanno mostrato che il NMDA (100 microM)-evocato rilascio di DA [H (3)] &egrave; stato diminuito significativamente dalla precedente esposizione delle fette SN a trans-ACPD. Infine, denervazione parziale del percorso DA nigro-striatali con 6-hydroxydopamine (6-OH-DA) aumentato rilascio trans-ACPD-indotta di DA [H (3)], mentre sono diminuiti effetti inibitori trans-ACPD NMDA-evocato release di [(3) H] DA fette nigral. I presenti risultati suggeriscono che il dendritico rilascio di DA nel SN &egrave; regolamentato da mGluR attivazione. Tali nigral mGluR attivazione pu&ograve; produrre fronte gli effetti al rilascio basale e NMDA-evocato di DA in SN. Inoltre, tali effetti mGluR-indotta in SN sono modificati in risposta alla denervazione parziale del percorso DA nigro-striatali.

Un sistema superfusion fue utilizado para estudiar los efectos de ligandos de receptores (mGluR) de glutamato metabotr&oacute;picos sobre la liberaci&oacute;n de dopamina [3 H] ([3 H] DA) previamente tomados por rebanadas de la sustancia negra (SN) de rata. &aacute;cido dicarbox&iacute;lico trans-(+/-)-1-amino-(1S,3R)-cyclopentane (trans-CONTINU&Eacute;; 100 y 600 microM), un grupo I y II mGluR agonista, evoc&oacute; el lanzamiento de [3 H] DA de nigral rebanadas. Este &uacute;ltimo efecto se redujo significativamente por (2S,3S,4S)-2-methyl-2-(carboxycyclopropyl)-glycine (MCCG; microM 300), un antagonista de grupo II mGluR, o por la adici&oacute;n de tetrodotoxina (D-APV; 1 microM) al medio superfusion. &Aacute;cido D-(-)-2-amino-5-Phosphono-Valeric (100 microM), un antagonista de los receptores N-metil-D-aspartato o la presencia de mg (1.2 mM) en el medio superfusion no modific&oacute; trans-CONTINU&Eacute;-inducida por liberaci&oacute;n de DA de [3 H]. Spontaneous calcium oscillations in mushroom bodies of late stage pupal and adult Drosophila brains have been implicated in memory consolidation during olfactory associative learning. This study explores the cellular mechanisms regulating calcium dynamics in Kenyon cells, principal neurons in mushroom bodies. Fura-2 imaging shows that Kenyon cells cultured from late stage Drosophila pupae generate spontaneous calcium transients in a cell autonomous fashion, at a frequency similar to calcium oscillations in vivo (10-20/h). The expression of calcium transients is up regulated during pupal development. Although the ability to generate transients is a property intrinsic to Kenyon cells, transients can be modulated by bath application of nicotine and GABA. Calcium transients are blocked, and baseline calcium levels reduced, by removal of external calcium, addition of cobalt, or addition of Plectreurys toxin (PLTX), an insect-specific calcium channel antagonist. Transients do not require calcium release from intracellular stores. Whole cell recordings reveal that the majority of voltage-gated calcium channels in Kenyon cells are PLTX-sensitive. Together these data show that influx of calcium through PLTX-sensitive voltage-gated calcium channels mediates spontaneous calcium transients and regulates basal calcium levels in cultured Kenyon cells. The data also suggest that these calcium transients represent cellular events underlying calcium oscillations in the intact mushroom bodies. However, spontaneous calcium transients are not unique to Kenyon cells as they are present in approximately 60% of all cultured central brain neurons. This suggests the calcium transients play a more general role in maturation or function of adult brain neurons.

In Drosophila, nicotinic acetylcholine receptors (nAChRs) mediate fast excitatory synaptic transmission in mushroom body Kenyon cells, a neuronal population involved in generation of complex behaviors, including responses to drugs of abuse. To determine whether activation of nAChRs can induce cellular changes that contribute to functional plasticity in these neurons, we examined nicotine-evoked responses in cells cultured from brains of late stage OK107-GAL4 pupae. Kenyon cells can be identified by expression of green fluorescent protein (GFP+). Nicotine activates alpha-bungarotoxin-sensitive nAChRs, causing a rapid increase in intracellular calcium levels in over 95% of the Kenyon cells. The nicotine-evoked calcium increase has a voltage-gated calcium channel (VGCC) dependent component and a VGCC-independent component that involves calcium influx directly through nAChRs. Thapsigargin treatment reduces the nicotine response consistent with amplification by calcium release from intracellular stores. The response to nicotine is experience-dependent: a short conditioning pulse of nicotine causes a transient 50% reduction in the magnitude of the response to a test pulse of nicotine when the interpulse interval is 4 h. This cellular plasticity is dependent on activation of the VGCC-component of the nicotine response and on cAMP-signaling, but not on protein synthesis. These data demonstrate that activation of nAChRs induces a calcium-dependent plasticity in Kenyon cells that could contribute to adult behaviors involving information processing in the mushroom bodies including responses to nicotine.

Dissociated cultures from many species have been important tools for exploring factors that regulate structure and function of central neuronal synapses. We have previously shown that cells harvested from brains of late stage Drosophila pupae can regenerate their processes in vitro. Electrophysiological recordings demonstrate the formation of functional synaptic connections as early as 3 days in vitro (DIV), but no information about synapse structure is available. Here, we report that antibodies against pre-synaptic proteins Synapsin and Bruchpilot result in punctate staining of regenerating neurites. Puncta density increases as neuritic plexuses develop over the first 4 DIV. Electron microscopy reveals that closely apposed neurites can form chemical synapses with both pre- and postsynaptic specializations characteristic of many inter-neuronal synapses in the adult brain. Chemical synapses in culture are restricted to neuritic processes and some neurite pairs form reciprocal synapses. GABAergic synapses have a significantly higher percentage of clear core versus granular vesicles than non-GABA synapses. Gap junction profiles, some adjacent to chemical synapses, suggest that neurons in culture can form purely electrical as well as mixed synapses, as they do in the brain. However, unlike adult brain, gap junctions in culture form between neuronal somata as well as neurites, suggesting soma ensheathing glia, largely absent in culture, regulate gap junction location in vivo. Thus pupal brain cultures, which support formation of interneuronal synapses with structural features similar to synapses in adult brain, are a useful model system for identifying intrinsic and extrinsic regulators of central synapse structure as well as function.

Voltage-gated calcium channels containing alpha1 subunits encoded by Ca(v)2 family genes are critical in regulating release of neurotransmitter at chemical synapses. In Drosophila, cac is the only Ca(v)2-type gene. Cacophony (CAC) channels are localized in motor neuron terminals where they have been shown to mediate evoked, but not AP-independent, release of glutamate at the larval neuromuscular junction (NMJ). Cultured embryonic neurons also express CAC channels, but there is no information about the properties of CAC-mediated currents in adult brain nor how these channels regulate transmission in central neural circuits where fast excitatory synaptic transmission is predominantly cholinergic. Here we report that wild-type neurons cultured from late stage pupal brains and antennal lobe projection neurons (PNs) examined in adult brains, express calcium currents with two components: a slow-inactivating current sensitive to the spider toxin Plectreurys toxin II (PLTXII) and a fast-inactivating PLTXII-resistant component. CAC channels are the major contributors to the slow-inactivating PLTXII-sensitive current based on selective reduction of this component in hypomorphic cac mutants (NT27 and TS3). Another characteristic of cac mutant neurons both in culture and in whole brain recordings is a reduced cholinergic miniature excitatory postsynaptic current frequency that is mimicked in wild-type neurons by acute application of PLTXII. These data demonstrate that cac encoded Ca(v)2-type calcium channels regulate action potential (AP)-independent release of neurotransmitter at excitatory cholinergic synapses in the adult brain, a function not predicted from studies at the larval NMJ.

Much of our current understanding about neurodegenerative diseases can be attributed to the study of inherited forms of these disorders. For example, mutations in the presenilin 1 and 2 genes have been linked to early onset familial forms of Alzheimer's disease (FAD). Using the Drosophila central nervous system as a model we have investigated the role of presenilin in one of the earliest cellular defects associated with Alzheimer's disease, intracellular calcium deregulation. We show that expression of either wild type or FAD-mutant presenilin in Drosophila CNS neurons has no impact on resting calcium levels but does give rise to deficits in intracellular calcium stores. Furthermore, we show that a loss-of-function mutation in calmodulin, a key regulator of intracellular calcium, can suppress presenilin-induced deficits in calcium stores. Our data support a model whereby presenilin plays a role in regulating intracellular calcium stores and demonstrate that Drosophila can be used to study the link between presenilin and calcium deregulation.

Gran parte de nuestra comprensi&oacute;n actual sobre las enfermedades neurodegenerativas puede atribuirse al estudio de las formas hereditarias de estos trastornos. Por ejemplo, las mutaciones en los genes de la Presenilina 1 y 2 se han ligado a formas familiares de temprana aparici&oacute;n de la enfermedad de Alzheimer (FAD). Utilizando el sistema de nervioso central de Drosophila como modelo hemos investigado el papel de la Presenilina en uno de los primeros defectos celulares asociados con la enfermedad de Alzheimer, desregulaci&oacute;n de calcio intracelular. Demostramos que la expresi&oacute;n de tipo salvaje o presenilin FAD-mutante en las neuronas de la Drosophila CNS no tiene ning&uacute;n impacto en la reclinaci&oacute;n de los niveles de calcio pero dan lugar a los d&eacute;ficits en las tiendas de calcio intracelular. Adem&aacute;s, demostramos que una mutaci&oacute;n de la p&eacute;rdida de funci&oacute;n en la calmodulina, un regulador clave de calcio intracelular, puede suprimir los d&eacute;ficits presenilin-inducida en las tiendas de calcio. Compensatory changes occurring during presymptomatic stages of Parkinson's disease (PD) would explain that the clinical symptoms of the disease appear late, when the degenerative process is quite advanced. Several data support the proposition that brain-derived neurotrophic factor (BDNF) could play a role in these plastic changes. In the present study, we evaluated the expression of the specific BDNF receptor, trkB, in a rat model of presymptomatic PD generated by intrastriatal injection of the neurotoxin 6-OHDA. Immunohistochemical studies revealed a decrease in trkB expression in SN pars compacta (SNc) seven days after 6-OHDA injection. At this time point, no change in the number of tyrosine hydroxylase (TH) immunoreactive (TH-IR) cells is detected, although a decrease is evident 14 days after neurotoxin injection. The decrease in TH-positive cells and trkB expression in SNc was significantly prevented by systemic administration of Ifenprodil, a specific antagonist of NR2B-containing NMDA receptors. Therefore, an NR2B-NMDA receptor-dependent decrease in trkB expression precedes the disappearance of TH-IR cells in SNc in response to 6-OHDA injection. These results support the idea that a functional coupling between NMDA receptors and BDNF/trkB signalling may be important for the maintenance of the dopaminergic phenotype in SNc during presymptomatic stages of PD.

An NR2B-Dependent Decrease in the Expression of trkB Receptors Precedes the Disappearance of Dopaminergic Cells in Substantia Nigra in a Rat Model of Presymptomatic Parkinson's Disease.

Biogenic amines (BAs) play a central role in the generation of complex behaviors in vertebrates and invertebrates, including the fly Drosophila melanogaster. The comparative advantages of Drosophila as a genetic model to study the contribution of BAs to behaviors stumble upon the difficulty to access the fly brain to ask relevant physiological questions. For instance, it is not known whether the activation of nicotinic acetylcholine receptors (nAChRs) induces the release of BAs in fly brain, a phenomenon associated to several behaviors in vertebrates. Here, we describe a new preparation to study the efflux of BAs in the adult fly brain by in vitro chronoamperometry. Using this preparation we show that nAChR agonists including nicotine induce a fast, transient, dose-dependent efflux of endogenous BAs, an effect mediated by α-bungarotoxin-sensitive nAChRs. By using different genetic tools we demonstrate that the BA whose efflux is induced by nAChR activation is octopamine (Oct). Furthermore, we show that the impairment of a mechanically induced startle response after nicotine exposure is not observed in flies deficient in Oct transmission. Thus, our data show that the efflux of BAs in Drosophila brain is increased by nAChR activation as in vertebrates, and that then AChR-induced Oct release could have implications in a nicotine-induced behavioral response.

nAChR-induced octopamine release mediates the effect of nicotine on a startle response in Drosophila melanogaster.

Drosophila melanogaster has been successfully used as a simple model to study the cellular and molecular mechanisms underlying behaviors, including the generation of motor programs. Thus, it has been shown that, as in vertebrates, CNS biogenic amines (BA) including serotonin (5HT) participate in motor control in Drosophila. Several evidence show that BA systems innervate an important association area in the insect brain previously associated to the planning and/or execution of motor programs, the Mushroom Bodies (MB). The main objective of this work is to evaluate the contribution of 5HT and its receptors expressed in MB to motor behavior in fly larva. Locomotion was evaluated using an automated tracking system, in Drosophila larvae (3(rd)-instar) exposed to drugs that affect the serotonergic neuronal transmission: alpha-methyl-L-dopa, MDMA and fluoxetine. In addition, animals expressing mutations in the 5HT biosynthetic enzymes or in any of the previously identified receptors for this amine (5HT1AR, 5HT1BR, 5HT2R and 5HT7R) were evaluated in their locomotion. Finally, RNAi directed to the Drosophila 5HT receptor transcripts were expressed in MB and the effect of this manipulation on motor behavior was assessed. Data obtained in the mutants and in animals exposed to the serotonergic drugs, suggest that 5HT systems are important regulators of motor programs in fly larvae. Studies carried out in animals pan-neuronally expressing the RNAi for each of the serotonergic receptors, support this idea and further suggest that CNS 5HT pathways play a role in motor control. Moreover, animals expressing an RNAi for 5HT1BR, 5HT2R and 5HT7R in MB show increased motor behavior, while no effect is observed when the RNAi for 5HT1AR is expressed in this region. Thus, our data suggest that CNS 5HT systems are involved in motor control, and that 5HT receptors expressed in MB differentially modulate motor programs in fly larvae.
